DoesitmakeadifferenceifyourmousehasaB6J,B6N,ormixedB6J;B6Nbackground?Ifyouransweris“No,”considerthefollowing: C57BL/6JhastheNntmutation,whichmodulatesmetabolism(Freemanetal.2006;Shimomuraetal.2009)andimmuneresponse(Ripolletal.2012). C57BL/6JandC57BL/6Nvaryintheirfearresponses(Radulovicetal.1998;Stiedletal.1999)andhowanestheticsaffecttheircardiacfunctions(Rothetal.2002). C57BL/6NmiceandembryonicstemcellsderivedfromthemharbortheRd8mutationoftheCrb1gene,whichcausesdevelopmentaldefectsinphotoreceptorsegments(Mehalowetal.2003;Mattapalliletal.2012). MiceproducedbytheKnockoutMouseProject(KOMP)areonaB6Nbackground,whichmeansthatifyoubreedthemtoyourB6colony,youmaybeproducingoffspringwithamixedB6J;B6Ngeneticbackground. Whenyoureceivestrainsfromothersources,keepingthemutationonsimilarstrainbackgroundreducesthechancethatyouwillalterorloseyourstrain’sphenotypeduetobackgroundmodifiergenes. Onceyouknowthebackgroundofyourmousestrain,youcanbetteridentifytherightcontrolstrain,whichbackgroundtoselectforbackcrossing,andwhatstrain-specificphenotypesyoushouldbeawareof. Howyoucanusethe C57BL/6SubstrainCharacterizationPanel JAX’snewC57BL/6SubstrainCharacterizationPanel coversallautosomesandtheXchromosome.Thegenome-widespacingaverages15.4+/-7.3Mb–equivalenttoapproximately8centiMorgans.
