ThePrkdcscidmutationa,commonlyknownas“scid”or“severecombinedimmunodeficiency”,essentiallyeliminatesadaptiveimmunity.[5]Prkdcscidisaloss-of-functionmutationinthemousehomologueofthehumanPRKDCgene,whichencodesaproteinthatresolvesDNAstrandbreaksthatoccurduringV(D)JrecombinationindevelopingTandBlymphocytes.[6]MicehomozygousforthemutationhaveseverelyreducednumbersofmatureTandBcells.[3][5]ThephenotypicpenetranceofPrkdcscidvariesamonginbredstrainbackgrounds,butthemutationismosteffectiveateliminatingadaptiveimmunityontheNODgeneticbackground.[3]DuetothePrkdcscidmutation,however,theNSGstrainshowshighsensitivitytoradiation,T-cellleakage,andincreasedincidenceofthymiclymphomaformation;assuch,thisstraincannotbeusedtopredictclinicalresponsetocertainanticancerdrugs,orforlong-termtransplantationstudies.[7][8]
