AtleastsixautosomalcodominantallelesofthegeneDNASE1havebeencharacterized,DNASE1*1throughDNASE1*6,andthesequenceofDNASE1*2representedinthisrecord.Mutationsinthisgene,aswellasfactorinactivatingitsenzymeproduct,havebeenassociatedwithsystemiclupuserythematosus(SLE),anautoimmunedisease.[7][8]ArecombinantformofthisproteinisusedtotreatoneofthesymptomsofcysticfibrosisbyhydrolyzingtheextracellularDNAinsputumandreducingitsviscosity.[9]Alternatetranscriptionalsplicevariantsofthisgenehavebeenobservedbuthavenotbeenthoroughlycharacterized.[5] Ingenomics[edit] Ingenomics,DNaseIhypersensitivesitesarethoughttobecharacterizedbyopen,accessiblechromatin;therefore,aDNaseIsensitivityassayisawidelyusedmethodologyingenomicsforidentifyingwhichregionsofthegenomearelikelytocontainactivegenes[10] DNaseISequenceSpecificity[edit] IthasbeenrecentlyreportedthatDNaseIshowssomelevelsofsequencespecificitythatmaydependonexperimentalconditions.[11]Incontrasttootherenzymeswhichhavehighsubstratespecificity,DNaseIcertainlydoesnotcleavewithanabsolutesequencespecificity.However,cleavageatsitesthatcontainCorGattheir3'endislessefficient.
